Diospyros kaki Thunb cv.Fuyu and Jiro are Japanese persimmon varieties. Dormant buds taken from persimmon trees, as in vitro explants materials, were set up successfully. New shoots were attained. Callus was induced and adventitious buds were differentiated from leaves and roots of tissue cultures; new shoots propagation and shoots rooting were studied in different basal medium added different hormones. The paper could be usefull for modified gene persimmon and for fast-propagation plants as well. Results as following:1 Micropropagation of two persimmon verities was conducted. Effects of basal mediums and hormones on the propagation were studied by Orthogonal design trial. Results showed DKW medium was the best basal medium, and MS was much better than WPM which could not be used for the two persimmon propagation. 'Fuyu' persimmon in ZT 1.0 mg/L without BA medium grew best. There was mucfi more longer shoots in ZT 2.0 mg/L for 'Jiro', but callus was larger which was not beneficial to the micropropagation. DKW medium containing ZT 1.0 mg/L and BA 0.5 mg/L was suitable for the 'Jiro' micropropergation.2 Effects of basal mediums, hormones and their concentrations on shoots regeneration from leaves of 'Fuyu' and 'Jiro' were studied. TDZ was founded to be proper for bud differentiation, the bestshoot buds from leaves of 'Fuyu' and 'Jiro' induction media was 1/2DKW+TDZ 0.5 mg/L. After 30 days, the calli were subdivided and transferred to 1/2DKW medium supplemented with 1.0 mg/L ZT and 0.1 mg/L IAA.. After 30 days, regeneration rate exceeded 90%,and average No. of bud of each explant with buds exceed 7.8,5.6 respectively.3 New shoots were differentiated from roots of 'Fuyu' and 'Jiro' shoots after 20 days since culture in the medium containing ZT and average no. of shoots was 6.7, 6.3 respectively.4 When subculture times is less than 15, 1/2MS medium suits rooting of 'Fuyu' and 'Jiro' persimmon. 1/2MS and 1/2MS(1/2N) medium both suit rooting from 'Fuyu' and 'Jiro' shoot after subculture times is more than 15. IBA was more effective than IAA. Shoots cultured in medium containing IBA 1.0 mg/L and exceeding 90 percent of shoots are rooted.5 Regeneration shoots from roots have high rooting ability, but rooting ability of regeneration shoots from roots descended along with subculture times.
