Base on the morphological characteristics and genetic diversity, the populations of Medicago falcata from provinces of Liaoning, Inner Mongolia, Xinjiang and Tibet of China and former Russia were studied on genetic structure, genetic diverse and evolvement potential. The results are as follows1. There is higher rate of hard seed among the populations of Medicago falcata. The phase of seedling of the wild population is longer, and their height of seedling stage were shorter. The leaf length, leaf width and leaf shape are different in different populations. There is little different among the populations about D/F, but it is lower.2. Random amplified polymorphic DNA (RAPD) analysis demonstrated that 21 random primers produced 188 loci, obtaining 188 polymorphic loci in 6 populations. Polymorphic loci among the populations: Xinjiang > Liaoning >Xilinguole> former Russia > modern pasture> Tibet. By Shannon index evaluation, 28.05% of genetic variation was between populations and 71.95% of genetic variation was among the populations. The differentiation among populations was at a high level. Based on Nei index, the genetic differentiation between the populations was 0.2498, and a majority of genetic differentiation was among the population (0.7502). The Shannon index and the Nei index indicated that: Xinjiang> Liaoning>Xilinguole>modern pasture>former Russia>Tibet.Cluster analyses based on RAPD makers shows that the most genetic identity is between the population from Liaoning and the population from Xilinguole. And the most genetic distance is between the population from modern pasture and the population from former Russia. The genetic distance among the populations is 0.0714.3. Random amplified polymorphic DNA (RAPD) analysis demonstrated that the genetic diversity between the populations was at a high level (Gst=0.2498), and there are more genetic distance and the less genetic identity between the populations of China and the populations of former Russia. So we should use the germplasm from original habitat or from foreign.
