In this research, some egesta and puke samples from pigs have been done with and inoculated into the ST cells, they could reproduce on ST cell lines and produce prominence CPE: the infected cells appeared gathering and amalgamation. According to the nucleic acid type experiment and some other physical experiments we found the virus was a kind of RNA virus which with envelope, and took on positive in the neutralization experiment, electron microscope experiment were used to detect the virus and found the virus was about 90nm in diameter.In a word, one Transmissible gastroenteritis virus (TGEV) strain was isolated successfully from the puke, egesta and so on,we named it SC-A strain. According to these publiced gene sequencese in the GenBanK, one pair primers were designed by Oligo6.0 and amplied about 1.8kb fragment from the TGEV SC-A S gene, then clone the fragment into pMD18-T and sent to be sequenced. The whole sequence which is 1760 bp long.Alignment of pairs of sequence indicates that there are 98.8% a similary with other Transmissible gastroenteritis virus strains, respectively. Multiple sequence alignment discloses that there are a few difference between TGEV SC-A S gene and other TGEV genes. Alternative codon in TGEV SC-A-S have distinctly different frequenty by codon bias analysis at EMBOSS, there ishyd -rophobicity in TGEV SC-A protein. According to the anaysis of the antigen sites in the SC-A-S gene, there are some antigen sites in the SC-A S gene.
