In this study, BALB/c mice were immunized with 50μg , 100μg , 100μg +pcDNA-IL-2(100μg ),200μg pcDNA-PRRSV-SC2-ORF5 gene vaccine by the route of intramuscular injecting and 6μg gene gun bombardment. Injecting site muscle, cardiac muscle, Stomach, duodenum, liver, spleen, kidney, lung and brain of different groups were collected in different times post inoculation, distribution and expression were detected by PCR, ISH, ISPCR and immunohistochemistry , we had conclusions with these methods ①sensitive of PCR is higher than sensitive of ISH and ISPCR; ②sensitive of ISPCR is higher than ISH and gene vaccine could be detected in longer time BALB/c mice, at the same time, intension is higher;③IL-2 could protract preserving time of gene vaccine in BALB/c mice and the longest time was 30d;④There is positive relation of dose of gene vaccine and preserving time in collected tissues of mice of intramuscular groups. ⑤the dose of gene vaccine of gene gun bombardment group is tittle while gene vaccine preserving time is also long , ⑥ the dose of gene vaccine of gene gun bombardment group is tittle, but expression of detected gene vaccine is more than intramuscular groups of the same time and expression distributed in many tissues, all of which demonstrated that between gene gun bombardment and intramuscular injecting are better immune route while gene gun bombardment effect is better than intramuscular injecting. ⑦expression of intramuscular injecting of gene vaccine and pcDNA-IL-2 was more than intramuscular of gene vaccine only, which demonstratepcDNA. IL-2 is better adjuvant for ORF5 gene vaccine. ⑧there is positive relation of gene vaccine dose and expression of intramuscular injecting group.
