Black Rot is one of most important diseases which has caused heavy losses the sweet potato production in the world. Use of resistant culticars has been considered as an effective and important measure for preventing production from the damage caused by the disease.AFLP is a new effective method of molecular marker. It was mostly used on the research of high mammal and plants. The fingerprinting analysis based on AFLP was carried out in 12 different sweet potato coming from different areas.Identification of resistance to b sweet potato black rot in germplasm was conductod by using iniecting-inoculation method .Among 12 accessions,4 resistant, 3 tolerant and 5 susceptible.A most effective AFLP technological system was established . Some improved factors were below:restriction time was 3 hours; the most available condition of ligation reaction was 22℃,3 hours and 16℃,10 hours;preamplification reaction system was 50 ul;amplification reaction system was 20 ul.One hundred thirty-two pairs of AFLP primers were used to generate polymorphic bands and one specific DNA fragment was amplified by E06 / M07 primer combination in sweet potato Black Rot; The specific DNA polymorphic fragment, a 273 base pair band, was excised and cloned into T-easy vector. The results provide an useful aid to establish the system of sweet potato Black Rot. resistant breeding.
