| In this paper, we used the annual resting stem of different varieties of Vitis amurensis Rupr. as materials. We got the suitable media on the different stages and analyzed the factors in the plant and rapid propagation technology system. And at the same time we studied the Virus-free and micrografting technology of Vitis amurensis Rupr..The results of experiments were as follows: in the plant and rapid propagation technology system of Vitis amurensis Rupr., in the initiation culture stage, the suitable medium was B5+IAA0.2mg/L for "Zuo Shanyi", "Zuo Shan'er", "Shuang Hong", "Shuang You" and "Shuang Qing", B5+IAA0.5mg/L was the optimal medium for "Shuang Feng". During the course of multiplication and rooting cultivation, the suitable medium was 1/2MS+IBA0.3 mg/L for"Zuo Shanyi" and "Shuang Hong", reforming B5+IBA0.5mg/L was the compatible medium for "Zuo Shan'er" , "Shuang You" and "Shuang Qing", the suitable medium for "Shuang Feng" was l/2MS+IBA0.5mg/L.In the acclimatization the suitable base medium for six varieties was river sand. In addition, river sand: humus: sawdust =1:1:1, sawdust: river sand=2:l and sawdust: river sand= 1:1 was also suit for some varieties.During grape Virus-free culture, we selected the suitable medium for microapical culture:MS+6-BA2mg/L+NAA0.05mg/L+sucrose 30g/L+agar 9g/L.In the course of Virus-free treatment, the combination of tip culture and heat treatment can make tip survival rate obvious more than single tip culture. When tip length was 0.4mm and the time for heat treatment was 20-30d, tube seedlings growed well, death rate was almost zero and the tip survival rate was good.And at the same time, we studied micrografting technology of using Vitis amurensis Rupr. as rootstock and "Jing Xiu" as scion. We found the suitable sampling parts and methods for grafting. In eight micrografting methods, Vitis amurensis Rupr. cultured for 30-40d with or without root and without leaf was selected as rootstock, the shoot tip of "Jing Xiu" tube seedlings cultured for 20-30d was selected as scion, the survival rate , germination rate and rooting for grafting seedlings was high. The growth time of grafting seedlings was longer 5-8days than tube seedlings, the joint of grafting should emerge in the air when transplanting grape grafting seedlings. |
