GnRH (Gonadotropin-releasing hormone) is an important signal molecule in the three systems: nervous system, immunity system and incretion system. and it is also one of the most important hormones which play important roles in the process of animal's procreation.In order to probe into the rule of GnRH's distribution in Cherry Valley Duck's brain stem, 15 Cherry Valley Ducks were chosen to be experiment animals and immunohistochemical method was also applied to localize GnRH which was contained in the nerve cell of brain stem in this experiment. The 15 experiment animals were divided into three groups: young male group, young female group and mature female group. After one weeks' feeding under routine feeding condition, the experiment began. First of all, the Cherry Valley Duck was narcotized and was put on the operating table, then it's brain tissue was fixed with paraformaldehyde by using perfusion fixation. After that, using surgical instruments to shell the cranium and the brain was taken out and droped into formaldehyde solution to enhance the fixation. The fixed duck brain would be made into paraffin block after a series of procedure such as dehydration, clarity, waxdip and so on, then the paraffin block was sliced into sections characterized by the thickness from 5 to 6 microns. The sections were stained by 2 methods. One method is Hematoxylin-Eosin Staining. The location and conformation of nucleus in brain stem can be observed by using this methods. Another method is immunohistochemical method named SABC by which the GnRH-immunoreactive neurons in brain stem can be studied.The results of this experiment indicate that the neurons which contain GnRH weredistributed in the main nucleus in the brain stem from the diencephalon to medulla oblongata. In this dissertation, The rule of GnRH's distribution in Cherry Valley Duck's brain stem was discussed on the basis of morphology and some viewpoints were advanced. The viewpoints hold that the expression of GnRH in neurons in the Cherry Valley Duck's brain stem were different when the ducks were in different grown stage, but there is no apparent difference between male and female duck which are both in the same grown stage. The difference of GnRH's distribution in brain stem between Cherry Valley Duck and mammals was also be compared and we found that there was a huge difference between them base on the genus difference. In the end, ground on morphology and through studied the GnRH's distributing rule, the possible new way of GnRH releasing was also suppose: GnRH may infiltrate into the third ventricle directly through PVHT and play physiological functions.The results of this dissertation can fill up vacancy of the research on localization of GnRH neurons and GnRH's possible new releasing way in the poultry field, and also can offer some morphological data for the development of basic theory about procreation-incretion in poultry. Moreover, the result can be compared with other different animal or species' GnRH neurons in the deep research.
