Mei flower (Prunus mume Sieb.et Zucc.) is one of the ten famous traditional flowers in China favored by Chinese people. Among Mei flower cultivars, the double flower cultivars possessing ornamental value play an important role. In the study, the feasibility of constructing a molecular genetic map using "double pseudo-testcross" strategy was discussed through establishing a F1 population. To find markers linked to the target characters, we also do some research applying BSA method. The main results of research are as follows:A F1 population with 33 individuals was constructed from the cross P. mume 'Xue Mei' (single flower, single pistil) and P. mume 'Fenpi Gongfen' (double flower, multi-pistil). The RAPD markers segregating in a 1:1 or 3:1 ratio account for 28.57% of the total RAPD markers. Therefore, it is feasible to use "double pseudo testcross" strategy to construct molecular genetic maps of Prunus mume.The SSR, ISSR reaction systems were established. And the probability of MADS-box-like genes existing in Mei flower was primarily testified based on SSR result.Parentage determination of 27 individuals from the F1 progeny 'Xue Mei' x'Fenpi Gongfen' by RAPD markers was performed and 23 of them are hybrids.The heterozygosity of 'Xue Mei' and 'Fenpi Gongfen' is 22.73% and 28.57% respectively based on the segregation patterns of 28 RAPD markers in F1 population.Using BSA method by RAPD markers, a marker, S109-1500, associated to the target traits was found.
