In 1985, Yue xin-Liu firstly reported an active substance of small molecular weight distilled from healthy puerperal woman which was named after placental peptide, and the major component of which was proved poly-peptide through physic-chemical character assay and and bio-activity research. Pharmacological and clinical experiments have proved that it has extensive immunological and pharmacological activity. But the limited number of human placentas and the appeared ethnics promote that researchers more and more attach importance to research and apply animal placentas. Goat placental immunoregulating factor is a poly-peptide of small molecular weight distilled from healthy puerperal goat. In order to evaluate the effects of GPIF on non-specific immunologic-regulating function in mice, animal pharmacological experiments have been carried on. In vitro tests, the normal macrophages and immunodeficiency macrophages caused by 0.1 mg/ml Cyclophosphamide were cultured in RPMI-1640 culture medium which contain GPIF concentration of 1,0.5,0.25,0.125,0.0625mg/ml in experimental groups, and RPMI-1640 negative control group and 0.1 mg/ml thymosin positive control group were set up. By the end of cell culture, the phagocytic ability of peritoneal macrophage to neutral red,reduction ability to MTT, and the amount of NO andIL-1 were measured, and variation were compared. In vivo tests, the animal model of immunity-suppressive murine were set up by 60Coγradial irradiation in experiment groups, every murine was IP injected GPIG concetration according 25,12.5,6.25mg/kg every day, and modle group,normal negative control group and 10mg/ml thymosin positive control group were set up. Every murine have been administered GPIF for 7 day, the phagocytic ability of peritoneal macrophage to chincken RBC, ability of macrophage carbon clearence were detected,and variation were compared. The results showed that, in vitro tests, Compared with control groups, GPIF could significantly augment the peritoneal macrophage phagocytic ability to neutral red, also can notabily improves the reduce ability of macrophage, and increases the amount of NO and IL-1 excreted by peritoneal macrophage; In vivo tests, GPIF significantly augment the peritoneal macrophage phagocytic ability to chincken RBC, each experimental groups also could notably improve the clearence ability of immunity-suppressive murine. GIPF possesses notable effects of non-specific immunologic enhancement to murine.
