Salinization of soil is a severe problem that has negative effects on farm production and ecological environment. The yield of soybean is severely affected by soil salinity. Enhancing the ability of soybean to survive salt condition is very important to improve the yield of soybean and the utility of the salinity soil. Much attention is paid to raising salt-tolerant plants, studying salt-tolerance mechanism,locating and isolating salt-tolerance genes. The purpose of this study was to map the salt-tolerant gene and create condition for molecular assisted selection in soybean. Three crosses of salt tolerant cultivar with susceptible cultivar were used as materials in this experiment. A resistant and a susceptible DNA bulk were created by pooling the DNA of 15 resistant and 15 susceptible F2 plants from the Wenfeng7 x Union F2 population, Jingdou33 x Hark F2 population, Tiefeng8 x Zaoshu6 F2 population respectively, Contrasting DNA bulks were screened with 422 pairs of simple sequence repeat (SSR) markers. We identified 10 marker that was related to salt tolerant gene in three population. The salt tolerant gene was located between satt339 and satt255 on molecular linkage group N, the genetic distance and position between marker and the gene is different among the three cross, which indicates that the number of population, the difference of genetic background between the two parents and random sampling error may affect result gene mapping. So , it is important to select suitable population to gene mapping. We designed 52 pairs of primers from salt-related gene and EST from Genbank. Those primers were detected in DNA and RNA that abstracted in Wenfeng 7(salt-tolerant) and Union(salt-sensitive). We identified 6 pairs of primers that related to salt-tolerant gene and it can be used as reference to identify the salt-resistance of varieties. On the other hand, this study provided helpful trace about the molecular mechanism of salt-tolerance in soybean.
