The organogenesis research of woody plant is one of the most important problem in asexuality multiplication forest, walnut is an vital ecnomic cultivate.It is also one of the representative plant of woody plant for poor rhizogenesis.The research of rhizogenesis is important in both theoretics and practic .We first choose different woody plant ,according to these material Two-dimensional gel electrophoresis which is important in proteomics was improved.Some important steps,such as sample prepare, isoelectric focusing, balance , dyeing were improved.By comparing we find that when extract protein acetone deposition is better for leaves and stem.The method of trichloroacetic acid making powder is better for seeds.Colloidal coomassile brillant blue stained gel is a sensitive method. we also establish a technique method of 2-DE+MALDI-TOF-MS which applied in woody plant.Proteins expressed during rhizogenesis of walnut cotyledon were profiled with the techniques of improved proteomics.During rhizogenesis ( 5d rooting culture) ,44proteins were up-regulated,45 proteins down-regulated, and 108 protein maintained their level. For more study ,we found down-regulated proteins have more high molecular weight proteins;up-regulated proteins have more alkali and low molecular weight proteins.At last,7 proteins were chosen to make polypeptide...
