Cotton is the main crop and primary source of the natural fiber, and it is the second edible oil source plant in the world too. The abundant and variety classification make breeders to be able to operate and utilize thousands of gene of cotton, and then make it play an even more important role in human living. There are already many fundamental accumulation and development about genetic research of the cotton in domestic and international, including heredity and localization of some important shape and economic character molecular marker , construction of genetic map. It has already become the important subject for the scientific circle of cotton in our country that how to occupy a space in competing of cotton study in the future and promote its genomics research. It is the foundation and key that we construct cotton BAC library with independent intellectual property adopting the variety of our country. We can fish and analyze BAC clones which have important characters utilizing a molecular marker probe, and then find new molecular marker > isolate the important genes and launch functional and comparative genome research thoroughly. This have the signification and applied prospect for improving the heredity breeding research level of cotton in our country.The material used in this study was the zhongmiansuo12 suyuan7235 and Pima90-53. According to the method of Zhang H B and Peterson DG. a high-efficient technology system of BAC library construction was established. BAC libraries of the three cotton materials were constructed utilizing this strategy, taking the pIindigoBAC-5 as vector. BAC clone was stored in 384-well microtiter plates.The current BAC library of Zhongmiansuo 12 consists of 38,784 clones. A number of 179 BAC clones were picked at random indicated an average insert size of 120 kb with a range of 50 to 150 kb. Based on an estimated genome size of 2118Mb, the Zhongmiansuo 12 BAC library provides 2 haploid genome equivalent; The suyuan7235 BAC library comprises 30,336 clones. The insert size of clones varied between 50 kb and 150 kb with the average size is 120kb. The Suyuan7235 BAC library covers a 2 haploid genome equivalent; The Pima90-53 BAC library comprises 100,000 clones.
