Using Asparagus Officinalis L. UC800 as the material ,The paper erect a tissue culture of asparagus system, treat the asparagus test-tube plantlet with colchicine to get a polysomic individual,and to finding heredity and variation mechanism we have irradiated asparagus callus culture.1. Study about the undifferentiated tissue of the asparagus, the better explant for the forming of undifferentiated tissue is the twigs. the medium MS+ 4 mg/L NAA is the better one in the callus induced, and the medium MS+ 2 mg/L 6-BA+ 1 mg/L KT the better one in differentiation, the medium MS+ 2 mg/L IBA+2 % the better one in rooting.2 . Treat the asparagus test-tube plantlet with colchicine of differnt concentration in two different ways, to get a polysomic individual. The result shows that dealing with the twigs for 72 hours by smear method is the better option.the inductivity is up to 94 %,.doubling rate is up to 96.1 %.But the doubling rate is 84.4 % by soaking method.3. The callus of asparagus was radiated by 5 differnt does of 60Co-r rays form 1Gy to 20 Gy, and make different effect.(1) Corresponding to lGy, 5Gy, 10Gy, 20Gy radiation does, The variation of chromosome up to9. 4%, 36.5%, 31.2%, 48.2%.(2) The study identified the different does callus by RAPD. We selected 32 availability primers from 100 random primers, and produced 184 DNA bands. With statistica software make the result that 25 of them of polymorphism and the similarity coefficient of them are 0.912-0.983. The average similarity coefficient of them is 0.952.It is previously that the genetic resemblance is low as they are sharing common ancestry.(3) The genetic distance shows that the different characters of asparagus in proportion to the radiation doses.
