The male sterility of photo-sensitive and thermo-sensitive genic male sterile line of two-line hybrid rice fluctuates at low temperature, which makes the risk of two-line hybrid rice in seed production. The paper focuses on transformation of the tetracycline-regulated male sterile system into rice 4008s and Taibei 309(Oryza sativa L. sbuspjaponica Kato), in order to gain a new two-line hybrid genic male sterile line of which the male sterility is affected simutaneously by tetracycline and photo-thermo factors. When the new male sterile line meets abnormal low temperature at thermo-sensitive stage, the spraied tetracycline can penetrates into plant cell and binds to TetR protein, which makes the TetR protein departure from the operator of barnase gene. The expression of barnase gene will elimilate the fluctuation of the male sterility. It is expected for offering a safer technique approach for the two-line hybrid rice. The main results of the thesis are summarized as below:1. Genotype is a dominant factor in regeneration system of rice.2. The quality of calli can be improved by supplement of 1 mg/L absisic acid(ABA),including mitigation of callus browning.3. The callus introduced by young panicle is better than that of mature seed embryo.
