The study was mainly to select a correct, simple and quick determination method of amylose content of wheat-grain. The method was mainly used in identification of the waxy wheat, no-waxy wheat and their segregating generation and used in study of amylose accumulation of different types.3 varieties with different quantity of amylose types were selected in this study. In the study of half-grain determination way,studied the effect of different quantity of acid and I2 and in different time of stain on the OD. we compared the AC value that was measured in different the proceeding methods of wheat grain. The quantity of amylose was compared small group with half-grain and the method of half-grain with normal method. The method of half-grain was applied in determining amylose content of F1 segregating generation and in studying of amylose accumulation of different types. The result showed that: 1. The compared of the different quantity of acid and I2,the resulted showed : CH2COOH of 1.0ml1mol/L and the 0.2% I2 of 1ml was as the best reaction condition. On the condition, the starch-I2 complex that it with special color was formed, and the measure-result was better.The OD was can be affected by time of stain,and the OD was accurate after of stain 15min. 2. Comparing dense of NaOH, the quantity of NaOH ,time of gelatinization and heated and not heated of after gelatinization in the proceeding of grain , founding the result of grain-gelatinization was better when the dense of NaOH =1.0mol/L and the quantity of NaOH =1.5ml in proceeding of half-grain gelatinization. The result of grain-gelatinization was not significant with accession of dense of NaOH and the quantity of NaOH. 24h of gelatinization was the best time, if the grain was heated for 10min in boiling water bath after 24h of gelatinization, the amylose content was accurate. 3. The amylose content of half grain and small group was analyzed ,the correlation analysis illustrated r=0.989>r0.01=0.959,the r was very significant , so the amylose content of group could be supposed by the measured of half grain. T test was applied in the amylose content by the measured of half-grain method and normal method, two T–value that was measured by two methods was not significant, it (t =0.746
