Part I:The photo-thermo sensitive genic male-sterile rice is a kind of biological resources for the hybrid rice research. The proteomics technology now become a powerful tool for the investigation of the fertility in the present study. The 2-DE protein maps of rice F2(ORYZA SATIVA, peiai64S×9311) spike at meiosis stage, were obtained by optimizing crucial factors and procedures such as sample treatment, electrophoresis parameter, and gel concentration. The results revealed that adapting lysis solution to treating sample, choosing moderate amount of sample,using pre-cast immobilized linear dry strips for the first dimensional electrophoresis, 12. 5% separation gel and 4% concentrating gel were used to the SD S-PAGE electrophoresis.The 2-DE protein maps of the rice spikelet during the sterile and fertile stages were compared. 13 protein spots identified showed variations that were significant and reproducible , and searched their molecular function and biological process in the database. 74 Protein spots out of about 1000 detectable spots on the 2-D gels were identified by ESI-Q-TOF.
