| Porphyridium cruentum can synthesize some bioactive substances such as phycobiliprotein, polyunsaturated fatty acids (PUFA) and exopolysaccharide , which have application foreground. In this paper we investigated the metabolite regulation of active substances from Porphyridium cruentum; the effect of ultrasonic on the biology effect of Porphyridium cruentum; isolation and purify of phycoerythrin from Porphyridium cruentum, analyzed the stability of the phycoerythrin. The results were as follows:1. The culture conditions that affect the growth and metabolites production of Porphyridium cruentum were optimized with the uniform design.And DPS software was applied to get the optimum culture conditions for biomass , exopolysaccharide and phycoerythrin .As a resulte, the production had increased 140.4%, 98.9% and 133.6%,respectively. 2. The treatment conditions of the ultrasonic were optimized with the uniform design. And DPS software was applied to get the optimum culture conditions for biomass , exopolysaccharide and phycoerythrin .As a resulte, the production had increased 31.0% 54.3% and 15.9%,respectively. 3. Used fraction precipitation of ammonium sulphate, DEAE-Sepharose FF ion exchange column, B-phycoerythrin were separated and purified, the purity of the purified B-phycoerythrin reach 4.85 and a hydroxyl column , the purity of the purified B-phycoerythrin reach 5.10. 4. The physical and chemical properties of phycoerythrin from Porphyridium cruentum were experimentally studied and analyzed. B-phycoerythrin could be kept unlighted at the low temperature; Heavy metal and the acid condition have blight on B-phycoerythrin; Several usual food additives could keep the colour. |
