| The genetic relationship and genetic diversity of 20 phylogeography provenances of Fraxinus Manshurica Rup. in Northeast of China were studied by ISSR molecular marker technique. By analysing the growth charaters of 7 year-old F. Manshurica in three test sites, the provenance selection and district were brought forward.The ISSR molecular marker results were as follows:1. ISSR analysis of genetic relationship for 20 provenances of F. Manshurica carried out with 11 ISSR primers. 108 loci which containt 104 polymorphic loci were found and the percentage of polymorphic loci shared a range of 18.52%-37.96%. That of Provenance MSE was the highest (37.96%) and Provenance LSH was the lowest (18.52%).2. Genetic variation of F. Manshurica was counted in this paper with Shannon information index and Nei's gene diversity index. The range of Nei's index was from 0.1644 to 0.07590, and that of Shannon index was from 0.1103 to 0.2375. The variations of 20 provenances indicated by the two indexes were identical.3. The genetie diversity (Dst), based on Nei's method, among provenances of F. Manshurica was 0.1586. The coefficient of gene differentiation (Gst) is 0.6183. The estimate of gene flow from Gst (Nm) is 0.3086. There are 61.83% and 38.17% of total variation among and within provenances respectively.4. Genetic relationship cluster figure of 20 provenances of F. Manshurica was gotten based on ISSR marking technique. The provenances of F. Manshurica had been divided into 2 populations as follows. The first group contained provenance YH, DL, SCZ, MES, HL, LJ. FZ, HUA, HR, and ZH; and the second group contained provenance HUI, SL, DFH, TWH, WC, XF, XL, WQ, WDZ, and LSH. There's no significance of correlativity between genetic and geographical distance.Through investigating the growth charaters of 7 year-old F. Manshurica, geographical variances, the interaction of geographical variances and test sites, and the genetic stability of the 20 provenances were analyzed. The results of growth charaters analysis showed that:1. Among the provenances of F. Manshurica, there were significant difference in the high growth and there were no significant differences in root diameter growth.2. The height growth and stability were variable among provenances. The provenance SL was the best one in the both aspects and was selected as the best provenance.3. The interaction of provenance and site was significant. And the most suitableprovenance for specific test site was selected. Provenance blocks were defined. In Dailing test site, provenance YH, XL, SL, and LJ were the best. In Lushuihe test site, provenance HL, HUA, and SL were selected. And in Maoershan test site, we chose the provenance HUA as the most suitable provenance only. ?4. All provenances were organized into 4 blocks: They were South Mountin Changbai Block, Nouth Mountin Changbai Block, Block of Mountin Wandashan & Sanjiang Plain, Mountain Xiaoxing'an Block. |
