Recently, the eating safety of animal products has being gained more and more attentions by customers. Feed quality can affect eating safety of animal products and furthermore the development of animal husbandry. Aflatoxin B1(AFB1) in feed does harm to both animal and humans health, so it is great importance to determine accurately the content of AFB1 in animal feeds.Three main methodologies assay AFB1 content are TCL, HPLC and ELISA, among which ELISA is widely applied in feed industry due to its being simpler, faster and more accurate than the other two methods. But the accuracy of determination varies due to the complicated compositions in feeds. This study compared the accuracies of AFB1 determination by two protocols of ELISA, one of which was GB/T17480—1998 released by the central government, one was the modified protocol based on GB/T 17480—1998. One more step, this is, the extraction of AFB1 in organic phase was added to the GB/T17480—1998 protocol. The results showed that the modified ELISA protocol is more accurate than GB/T17480—1998 protocol. The accuracy was improved by 10% to 60%. 150 samples of animal feeds were analysed for AFB1 content by using the two ELISA protocols. The results indicated that AFB1 content in the feeds was overestimated by 20-80% when GB/T17480—1998 protocol was used. It is concluded that the modified ELISA protocol for AFB, determination is superior to GB/T17480—1998.
