In this study,seeds and stem segments of guayule were used as explants to establish a rapid micropropagation technique system in Guayule (Parthenium argentatum A. Gray) ,and especially to find out some effective measures to control the vitrification.The results were as follows:1)Seeds were the best explants considering the height of plantlet,the rate of contamination,vitrification and induction.2)The best initiation medium was l/2MS+00.01mg/LIBA.3)The best medium for multiplication was MS+BA1.0mg/L+KT0.5 1.0mg/L+ NAA0.1mg/L+IBA0.05mg/L,in which the multiplication rate was 14.33.4)The best medium for the rooting of shoots was l/2MS+IBA0.05mg/L+ AgNO30.5 mg/L+AC0.2%.5)For transplanting of plantlets,the best medium was rotten manure 10%+fertile soil30%+sand60%, the survival rate of plantlets being 63.33%.6)The best medium for plant regeneration from leaves was MS+BA0.5mg/L+ KT0.5 mg/L+IAA0.1mg/L,where the inductive rate was 81.94%.7)Methods to control vitrification:â‘ The seeds as explants could reduce the occurrence of vitrification.â‘¡Increasing the content of agar to 10g/L in the medium could control vitreous shoots,and the rate of vitrification was 2.38%.â‘¢Adding penicillin at 5mg/L to the medium could reduce the rate of vitrification to 2.98%.? Considering vitrification and the status of plantlet growth,adding activated charcoal(AC) at 0.2% to the medium could reduce the rate of vitrification to 8.63%.?Reducing the number of shoots in a bottle could decrease the vitrified shoot proportion.?The vitrification rate occurred when using cotton plugs wrapped in cheese cloth was the lowest among all seal materials,in comparison with polyethylene films.
