Repeated immunization of normal BALB/c mice with separated thymocyte from mature duck result in the production of antibodies which react with duck thymocyte surface antigens. Five monoclonal antibody-producing hybridomas (B6.B8.C12.E6.H5) were isolated from immunized BABL/C mice showing this property. During the 8 weeks of the passages, the five hybridomas were tested for their stability of secreting antibodies, the result indicated that the five hybridomas have the abilities to secreting antibodies stably.In the immunohistochemistry experiment, tracing the originating and distributing of lymphocytes which reacted with monoclonal antibodies in duck spleen, thymus, and bursa of different development stages. The result indicated the four monoclonal antibodies (B8.C12.E6.H5) had resembling characteristic of reactivity which were that, a large proportion of lymphocytes were stained in the medulla of thymus of 8 week-old duck, but densely packed small lymphocytes were unstained in outer cortex of thymus. The same result occurred in thymus of 1 day-old, 1 week-old and 16 week-old ducks. However a little lymphocytes were stained in the medulla of thymus of 24 week-old duck. There were distributing strained lymphocytes in the cortex and medulla of the bursa lymphoid follicles and in the propria of the bursa. The stained cells belonged to different populations, one was a little lymphocytes in the cortex and propria of the bursa; the other was relatively large cells with irregular morphology and secretory material in the cortex and medulla of the bursa, there had no change in the development of bursa. There were several lymphocytes and dendritic cells stained in the germinal centers of duck spleen, obvious positive results occurred in the outer of the germinal centers. The plasma cells and erythrocyte were unstained in the splenic red pulp. Most lymphocytes were stained in the periarterial lymphoid tissue and the periellipsoidal lymphoid tissue. Monoclonal antibody B6 reacted with lymphocytes in the thymus as well as to the former's reactivity. But in the bursa most lymphocytes and epithelial cells were stained, a part of lymphocytes were stained in the germinal centers of duck spleen.The four monoclonal antibodies (B8.C12.E6.H5) reacted with the common cell surface marker of between mature T lymphocytes and dendritic cells. Monoclonal antibody B6 reactedwith the common cell surface marker of mature T lymphocytes, B lymphocytes and epithelial cells.All the work provided a certain reagents for the research in separated T lymphocytes, defined lymphocyte populations and subpopulations and explored biological function of lymphocytes in duck immune system, also provided reference of the originating and distributing of immunocyte in duck.
