The Study On Artificial Diets Of Rice Stem Borer And Primary Choice On Resistance To Bt Of Rice Stem Borer

With the development of modern biology and science technology and improvement of study of modern entomology, the study of artificial diet on insects is required. The improvement and design of artificial diet of rice stem borer are much needed. Based on artificial diet of soybean flour, nine new kinds of artificial diet are designed of which three better kinds of artificial diet are selected out. Meanwhile, fifteen components of artificial diet are selected from reported diet .The orthogonal design of fifteen factors and two levels is made. At the same time, fifteen components are tested and five key factors have been selected out .Finally the second orthogonal design of five factors and four levels is carried on and the new optimum component of artificial diet of rice stem borer is reached. In addition, the primary choice of resistance to Bt of rice stem borer is conducted through artificial diets of rice stem borer. The results are as follow:1. There is no obvious difference between rice stem borer reared by three kinds of artificial diet and that by fresh rice stem. Hence three kinds of artificial diet are all better.2. Component of artificial diet â…¡ is composed of artificial diet â…  and water-oat stem flour. The experiment result indicates that water-oat stem flour can obviously improve the pupation rate and oviposition.3. Component of artificial diet â…¢ is made up of artificial diet â…  and rice stem flour. The test shows that artificial diet â…¢ can improve the pupation rate of rice stem borer and prolong the survival days of the first generation larva, but have no difference with contrast. In second generation, the result is contrary.4. The results of the two orthogonal designs of artificial diet of rice stem borer both indicate that sucrose and wheat germ flour are key factors of survival days of rice stem borer. Rice stem flour, wheat germ flour, yeast flour and rice stem chaff flour affect the survival. Rice stem flour affects the mean pupal weight. On whole consideration, five components of artificial diet are important to development of rice stem borer and are five key factors.5. Through two orthogonal designs, the optimum component of artificial diet of rice stem borer is: rice stem flourl8.75g, soybeanl5g, wheat germ flour15g, rice chaff flour6.25g, water-oat flour7.5g, casein10g, yeast flourl8.75g, cellulose 7.5g, sucrose 15g, glucose 7.5g,vitamin C7.5g, vitamin B3g, cholesterol 0.375g, chlorination0.25g, Beck mix salt 2.5g.6. Rice stem borer resistive to Bt is not selected out by rearing with high concentration Bt protein in short time and low concentration Bt protein in long time.
