Analyzing Of The ESAg Of The Larvae In Muscle Of Canine Trichinella And Establishment Of The LAT

The trichinelliasis is caused by the trichinella spiralis which belongs to Order Enoplida, Family Trichinellidae, Genus Trichinella. Its prosopon parasitizes small intestine and its larva parasitizes muscle. It is a kind of Amphixenosis. In order to seek a diagnosis antigen that has the high sensitivity and the good specificity, we have analyzed the ESAg and the polypide antigen by SDS-PAGE and Western blotting. We have established the LAT and the IHA using the ESAg.The analyse of collected ESAg in vitro by SDS-PAGE, there were two transparent bands, which have the molecular weight of 45kD and 49kD. The analyse of the polypide antigen by SDS-PAGE, there were five bands, which have the molecular weight of 28kD, 36 kD, 43 kD, 53 kD and 49kD. The analyse of the ESAg by Western blotting the molecular weight of 49kD was differential. The molecular weight of 36 kD and 43kD about the polypide antigen was differential.In order to compare the immunity of the ESAg and the polypide antigen, we have established the LAT and the IHA using the ESAg and the polypide antigen. We have determined the suitable reaction condition and detected the 76 shares canine blood of Yanbian in this experiment. The results showed that the positive rate was 18.42% by the LAT using the ESAg, the positive rate was 15.79% by the IHA using the ESAg, the positive rate was 11.84% by the LAT using the polypide antigen. This indicated that the LAT was better than the IHA, the ESAg was better than the polypide antigen.For understanding the rise and fall of the antibody against Canine Trichinella ,we have detected the canine of the artificial infection. The results showed that the antibody could be detected on the 7th day after the artificial infection, the average antibody titre was the highest on the 50th day. The average antibody titre gradually decreased on the 67th day and achieved the 1:32 on the 121st.This indicated that the ESAg had a kind of the high sensitivity and the good specificity. It may be the developed antigen in penetrating study.
