Avian influenza (AI) is a serious disease of poultry throughout the world. Hemagglutinin (HA) is the major surface glycoprotein of AIV. It is the main target antigen in eliciting humoral immune response against AIV. Studies have showed that the antigencity and immunogenicity of HA 1 is greater than that of HA2.In this study, HA1 gene was amplified by RT-PCR, ligated into pMD 18-T Simple vector and then transformed E.coli. Gene sequencing analysis showed that the cloned HA1 is 1035bp in length. This gene shares 91.22%-99.52% homology with those genes published in GenBank.The HA1 gene was inserted into the expression plasmid pGEX-6P-l. The recombinant fusion protein was expressed in E.coli induced by IPTG in the form of inclusion bodies. The molecular weights of GST-HA1 is 63kD as demonstrated by Western-blot using AIV antibody against H7 subtype. These results indicated that this recombinant preotein has good antigenicity.
