The objective: To study on the Function- mechanism of Chinese traditional medicine of adjuvant Vaccine and use two bioactive compounds of invigorating vital energy .We aimed to investigate a new and economic adjuvant of FMD vaccine,the adjuvant could heighten the protective power of Current-common FMD vaccines and reduce side-effects of Current-common FMD vaccines,it could play down the ratio of FMD and lessen the expenditure of labor. The adjuvant could activate the cellular and humoral responses without compromising long-term immunity, at the same time we were able to offer a test method of studying adjuvant and the experimentation had a good investigative meaning.The representative were Astragulus Polysacharin and GinsenosideRbl,the primary bioactive compounds in Astragalus and Ginseng respectively. Firstly, observing the antibody liter's change of mice and pig given Astragulus Polysacharin;GinsenosideRbl and directly mixed bacterin of FMD by intramuscularly injection. The results: the difference of antibody titer which mice given once immunity compared with control was not marked HAlog2<7 (p>0.05) ,the difference of antibody titer which mice given twice immunity compared with control was best marked HAlog2<7 (p<0.01);the difference was best remarkable when mice were immunised by APS (0.5 mg/each) +vaccine,APS (0.1 mg/each) +vaccine and GinsenosideRbl (0.01 mg/each) +vaccine (p<0.01) ,the difference was remarkable when mice were immunised by APS (0.05 mg/each) +vaccine and GinsenosideRbl (0.002 mg/each) +vaccine (p<0.05) . The effective antibody titer didn't achieved when pig were immunised once (HAlog2<7) ,the antibody titer was far higher twice than once (HAlog2>7) in a word.The Astragulus Polysacharin and GinsenosideRbl can improve antibody of FMD,along with the increased concentration of the two Chinese traditional medicine's ingredients the antibody of FMD is increasing.Subsequently, Intestinal Mucous Membrane Microvascular Cells (IMECs) were cultivated. IMECs were identified via F-VIIIRagAb and silver nitrate, MVECs were proved that F-VIIIRagAb was masculine,MVECs had their characteristic F-VIIIRagAb, IMECs were dyed by silver nitrate, atramentous silver nitrate aggraded in the brim of MVECs namely intercellular space, taking on brown yellow,cells connected sawtooth and incarnated typical polygon.Finally, When Astragulus Polysacharin,GinsenosideRbl and vaccine Simultaneity acted on MVECs and keeped on culturing for a period of time we looked into the change of NOJL-6 and TNF- a .The result: when the vaccine of FMD (0.1 u L/mL) is used by oneself,the secretion of NO,IL-6 and TNF- a of MVECs is highly marked more than control group (p<0.01);however,when MVECs is swinged in with the Astragulus Polysacharin(450 u g/mL) and GinsenosideRbl(40u g/mL) adjuvant Vaccine of FMD,the secretion of NOJL-6 and TNF- a of MVECs is decreasing comparing with vaccine of FMD group and higher than control group (p<0.05 IK p<0.01) . The outcome indicated Astragulus Polysacharin and GinsenosideRbl all heightened the antibody of FMD and the two Chinese traditional medicine's ingredients could protected MVECs and avoided MVECs's damnification.
