"Mopan" persimmon is the famous-brand specialty of Hebei province, fruits has richly the medicine and nourishment value, yet the persimmon plucks stage centralize, easily softening and producing area's inconvenience, cause the fruits fester gravely and cut down "Mopan" persimmon goods rate enormously. At the moment "Mopan" persimmon lays in yet with some tradition means like the house's stack up and shelf's store in the open air and certainly to freeze to lay in giving priority to, but this kinds of means don't cause the persimmon keep a long period of hardness , affecting persimmon lays in to preserve freshness stage and the sale stage. But the means to utilize the outward source gene to transform fixes a direction to reform the properties capacity of "Mopan" persimmon can resolves the problem radically.This study aims at problem of "Mopan" persimmon's store and researches the regeneration system and preliminarily establishing ACS gene's transformation system of "Mopan" persimmon .This supply substance together with technical support for "Mopan" persimmon breeding, reform and speedily spread . The main result is as follows:1 The regeneration system of "Mopan" persimmon has been established. This test display that the optimum method of leaf regeneration is adoping the two times's induction . First of all choose the leaf explants that were subculture for 20~30days inoculated the 1/2DKW+TDZ0.5mg/L+ZT1.0mg/L medium for 7 day's darkness treatment., and then shifts to among the l/2DKW+ZT2.0mg/L to induced the adventitious buds.The shooting perentage is 86% and adventitious bud is 3.7.2 The rooting system of "Mopan" persimmon has been established . Two means of appending hormone directly and dip hormone was adopted. The means of appending hormone directly compare IAA IBA with culture medium and darkness treatment, obtaining the optimum scheme that IAA excelled IBA;1/2MS (1/2N) excelled MS(1/2N )and 1/2MS. The rate of rooting is 93.5% and roots per shoot is 4.7. Optimum dip in the IAA that the scheme is 250mg/L steeps dipping in 15~20min. The rate of rooting is 75% and roots per shoot is 1.8.3 Preliminary establishing genetic transformation. The optimization transforms means was fixd . The optimum bactericide which choose and selects is dose separately in the interest of Cef500mg/L and Sp50mg/L. Some resistance plants had been screened. The part of resistance plants were identified by PCR amplification, only 2 plants were preliminarily proved that ACS gene had been integrated into their genome.
