Primary Investigation In Rice Quality-related Genes Identification And Their Application In MRNA Level

In the study, rice endosperm library with its bioinformatics database, 274 different rice endosperm total RNA from Jiaxing Academy of Agricultural Sciences and seeds amylose content database were used.Firstly, the 10,382 ESTs in cDNA library of rice endosperm at our prophase work were analysed with bioinformatics, result shows there were altogether 14 highly-expressed genes. Of them, most encoded quality-related genes including storage proteins, starch, photosynthesis et al. Of them, 14 highly-expressed specially in library include glutelin C, 10 kDa prolamin precursor, 11S globin, 12s globin et al. It was exammined glycine-rich protein that was related with cell wall scathed and repaired-related proteins in the forepart of seed development.Secondly, in order to analyze genes expression profiles of 5 different kinds of rice tissues and to find the highly-expressed genes in endosperm, was performed by Cyber-T statistical framework. It was shown that quality-related genes and recovery genes higher expressed in endoperm comparing other tissues. Quality-related genes are storage proteins, allegic proteins and selenoprotein, Manganese efficiency related protein. Highly-expressed allergic proteins include seed allergenic proteinRA14, RAG2 and Bet v I allergen-like;pollen allergen Bet v 1-L origined from birch and β-expansin la precursor are all up-regulated in embryo. Recovery genes include Thaumatin, peroxidase, PR-5, LTP 5, Receptor protein kinase-like gene, RAB24 which were related with resistance, senescence-related gene, dehydrated menace induced gene, desiccation-related protein, serine proteinases, Calcium binding protein-like which all correlated with defense. Metallothionein-like protein 2,3, hot shock protein, Cysteine proteinase inhibitor-I are up-regulated in embryo. And annotated endosperm specificially highly-expressed genes mainly were storage protein, seed allergic protein RAM, metabolize genes, GBSS I , Constans-like gene;annotated embryo specificially highly-expressed genes mainly were Thionin, 16KDa oleosin.Thirdly, we chose allergic protein, lipid protein, selenoprotein, starch-related genes for labling to probe into quality-related expressed profiling and filter high quality rice classes. Result shows diversity among the eight genes is less, but the trend is different. The pattern between clone H117E12 and H073F05 is close, and they are far off Wx maturing transcription;the clone H147A06 annotated seed allergic gene is far off other two allergic gene whose pattern is close. Allergic genes, selenoprotein, lipid protein expressed weakly in about half of samples, seed allergic gene expressed obviously in majority of samples. Wx maturing transcription expressed obviously in Indica,weakly in Japonica;Wx immaturing transcription expressed unobviously.Fourthly, we chose twenty samples which expressed lowly in 3 allergic genes and expressed highly in lipid protein and selenoprotein. In all, there are two kinds of Indica including 3k4 and 2c25, thirteen Japonica including dank3 et al, five kinds of sticky rice including dangao2 et al. Then, these samples need testing pass field breeding to filter samples yielding highly,it can provide research measurement inresearch.Fifthly, we deduced pattern of Wx" and Wxb in rice. Wxa work only and Wxb don't work at all;Wx" and Wxb work together, Wxb work mainly;Wx" don't work at all. It shows genetic effect is determined mainly by Wx", Wxb is the reason that cause middle and low AC lower, but other mutation cause AC of sticky rice lacking. We find that Wx has definite relativity with AC, and sbel also has relativity with AC to a certain extent.
