| Antisense phospholipase Dγ(PLDγ) gene was introduced into Lankao906 wheat mediated by Agrobacterium. 14 tansformants were acquired by kanamycin selection ,PCR and PCR-Southern technique analysis. The test results showed that anti-PLDγgene was integrated into wheat genomes . The variance between transgenic plants(lines) receptor on the expression in salt resistance, drought tolerance, Powdery mildew resistence and agronomic characters were studied, and main results were as follows:(1) The transgenic wheat of anti-PLDγgene were obtained. the result of molecular test showed that a 430bp fragment was amplified from variant plants by PCR analysis,which accorded with the result of PCR-Southern analysis, so we primarily confirmed that anti- PLDγgene had been integrated into Wheat genome.(2)By salt resistence test, 2 tansgenic plants from xi-rich wumai acquired had more tolerance than the untransgenic plants.Transgenic lines f-2 and f-4 grew normally on the medium with 1.4% NaCL, and the seedling height and the lenghth of root apartly were 1.0cm,1.8cm and 0.5cm ,1.0cm. The test result was same with that of relative electronic conductivity of transgenic plants and receptor under 1% NaCl stress for 72 hours.(3) Dought-tolerance test demonstrated that transgenic lines p094 was enhanced to some extent. Compared with receptor, the tansgenic lines could grow normally on the medium with 475mmol/l mannitol, and the number of side root increased. The result corresponded with the result of relative... |
