In this paper, The AFLP system in Ginkgo bilola was established, 39 male Ginkgo germplasms from the United States, French, Japan, Holland and China were studied for genetic diversity analysis of male Ginkgo germplasms based on AFLP technique for the first time in China.Genomic DNA was extracted with an improved CTAB (cetyltrimethyl ammonium bromide) method. Both the ultraviolet absorption test and agarose gel electrophoresis test indicated that the quality of genomic DNA was high pure, and they were fit for AFLP analysis. Factors affecting the AFLP analysis system in Ginkgo bilola were studied, and an optimized AFLP system was established. The important factors in the system are:①quality of template DNA.②300ng template DNA should be used in the digest system with the reaction time is 6 hours.③the time of ligation should be 10 hours at least.④the optimum dosage of microlitre ligation products is 1μl.⑤the products of preamplification are diluted to 70 times with 1 XTE for selective amplificationWith 7 pair of primers combinations, genetic diversity of 39 male Ginkgo germplasms was assessed by AFLP. A total of 1242 bands were generated, including 1219 polymorphic bands. The polymorphic percentage was 98.2%, and the identification percentage per primer combination was 100%.①Genetic diversity analysis of domestic and foreign male Ginkgo germplasms.The results showed that genetic diversity of population in China was higher than that from foreign countries. PPB of France, USA, and China three populations ranged from 39.22% to 92.52%, Nei's genetic diversity varied from 0.1402 to 0.2167, Shannon's Information index ranged from 0.2095 to 0.3462, the maximum existed in China, and the minimum existed in USA. The total genetic diversity of three populations was 0.2251, the genetic diversity within population was 0.1706, and coefficient of gene differentiation was 0.2382, which indicated that 23.82% genetic variation existed among populations.②Genetic diversity analysis of male germplasms from different provinces.Among Guangxi, Guizhou, Zhejiang, and Shandong populations, genetic diversity of Shandong population was the highest, the PPB, Nei's genetic diversity and Shannon's...
