Pineapple (Ananas comosus) is one of the most widely grown fruit crops in the tropics and subtropics. Since the first application of the technology of tissue culture on pineapple in 1970's, great progress has been made in this field.In this paper, we described the establishment of the regerenation and gentic transformation systems of commercially important pineapple cultivar Tainong-16. Pineapple plants were obtained via two different regeneration ways in this work, one from cllus induction and the other from buds directely. It is found that the highest regeneration ratio can be obtained in liquid MS medium supplemented with 2.0 mg/l 2,4-D, 1.5 mg/l 6-BAand 0.1 mg/l NAAat pH4.0.To establish the genetic transformation system of pineapple cultivar Tainong-16, three different methods were tried in the experiments using a plant expression vector carrying gus reporter gene. The sonication-mediated transfer method gave the highest transient exprssion of gus gene, but no transformant could be obtained finally. No ransformant was obtained via Agrobacterium-mediated method. Transgenic pineapple plants could be obtained when the microprojectile bombardment -mediated method was used. Factors affecting the transformation efficiency in the microprojectile bombardment-mediated method wre further optimized for efficient transfer of foreign genes into pineapple.In this work, two plant expression vectors harboring the peanuts resveratrol synthase (RS) gene were constructed under the control of CaMV35S promoter and used for genetic transformation of pineapple. Six transformants were found have had RS gene intergrated into their genome by PCR analysis. Among them, transcription of the foreign RS gene was clarifird in two transformants by RT-PCR analysis.
