Magnaporthe grisea is a well-known ascomycete that causes rice blast. A better understanding of the molecular basis of this disease is beneficial for rice blast control. As a model of fungal pathogen, M. grisea has typical infection processes, including conidiation, germination and appressorium differentiation. Isolation of functional genes of M. grisea is helpful to the understanding of other fungal-plant interactions. The pre-penetration processes of M. grisea, especially appressorium differentiation and formation, have been well studied, while the post-penetration processes which is essential for infection cycle of M. girsea, including hypha differentiation and colonization, still left much to be understood. In this study, MgKINl gene from Kinesins in appressorium formation was cloned, its function and expression pattern was analyzed by targeted gene replacement and eGFP fusion, respectively.A microtubule-associated mechanical adenosine triphosphatase, a novel mechanochemical protein called kinesin, is found in squid axoplasm, optic lobes, and in bovine brain. Its unusual property of forming a high affinity complex with microtubules in the presence of a nonhydrolyzable ATP analog, adenylyl imidodiphosphate,while adding adenylyl imidodiphosphate or KCl,they were separated quickly.Kinesin uses the energy of ATP hydrolysis to move membraneous organelles along microtubules toward the plus end of the microtubule. While in neural axons,it forms a crossbridge between membraneous organelles and microtubules.In this study,as structural proteins moving organelles in appressorium formation,their function was important for appressorium formation.The results are listed as followed:1. Five sequences were found by keywords "Kinesin" from ESTs of M. grisea , Mag3391745 was selected,named as MgKINl ,for its highest score bits and lowest E-score.The full sequence of MgKINl was cloned referring to Kinesin EST BLAST results, including promotor region and open reading frame, with a size of 7074bp.It...
