The development of female gonad was mainly focused on. The study includes three parts of different levels: the histology research of the oocyte development of ovary, the research of yolk during the development of ovary and the physiological changes during the development of ovary. The preserved ovaries were embedded in paraffin, sectioned at 6-8 μm thickness, and H-E stained. The mature development of oocyte was divided into five stages and the development of ovary was divided into six phases accordingly. The result shows that the pioneer ovary came into being at 41 days after fecundation. Most oocytes stayed at the second stage in August, the ovary was in the phase II, and started to develop since September . The spawn started from March to May of the next year. The ovary began to degeneration in June and when July came the ovary has been back to phase II, and most occytes remained was at the second stage. The mature of oocyte of seemed to be asynchrony and spawn repeatedly during the reproducing season. The content changes of the lipovitellin(LV) in female Odontobutis potamophila ( Gtinther ) was obtained by using the ELISA and Western blotting assay. The molecular weight of LV is 440KD. No LV was observed from July to September, the LV started to accumulate at October and reached the top at March the next year, still kept on a high level during April and May, then declined markedly. The contents of the water-solubility proteins show the similar trend while have some basic contents in July, August and September. The results indicates that the ovary of female Odontobutis potamophila ( Gtinther ) develops from October, which is beyond the result of morphology. The contents of estradiol in the female serum changed fluctuantly during the year. There were three fastigiums which occur in October, January and February, May. The expression of vitellogenin(VTG) mRNA in liver started from October and reached the top in March, and then the expression dropped. No VTGmRNA expression was observed since July. The VTG in serum changed similarly. The mechanism of how the estradiol regulates the expression of VTG needs further study. | Keywords/Search Tags: | Odontobutis potamophila (Gunther), oocyte development, histology, lipovitellin, yolk protein, fat, ELISA, Western blotting, RT-PCR, vitellogenin, VTGmRNA, estradiol | PDF Full Text Request | Related items |
