| Soybean (Glycine max) is the most important source of both edible vegetable oil and protein supplement for livestock. It also has a wide range of industrial uses. It is widely planted in our country. Improvement of soybean quality by genetic engineering is a more effective and fast way than traditional breeding. But to date, transformation frequency of soybean is still low relative to that obtained in other plants. To establish an effective transformation system is still a common aim of scholars.In this study, cotyledonary nodes from five varieties of soybean were used as the explants in the regeneration experiment. To establish a high frequency regeneration system for the gene transformation of soybean , the effect of seed germination time, explant size, germination culture medium, inductive culture medium and plant growth regulators were studied. The results show that seed germination time has effect to shoot regeneration, the best time is 5-7 days .The full cotyledonary is the best explant size for shoot differentiation. Different genetypes need different germination and inductive culture mediums, the best germination culture medium for Taiwan 292 is 1/2MS+1mg/L 6-BA and for E8157,Xiangchun 18, Xiangchun 13 and Xiangchun 15 is 1/2MS+2mg/L 6-BA. The best inductive culture medium for Taiwan 292, E8157 and Xiangchun 15 is MS+2mg/L6-BA,for Xiangchun 18 is MS+1mg/L6-BA and for Xiangchun 13 is 1/2MS+2 mg/L6-BA +0.05 mg/L IBA.
