Nitrogen is the biggest requirements nutrition factor in plant developmental process .It isn′t only the chemical composition of protein essential elements, but also is the composition of chlorophyll, enzymes and multi-vitamins.In life-sustaining activities and increase crop yields, improve quality of products, it is a crucial role.The formation of wheat seeds yield and quality is closely related to nitrogen nutrition.To clear impaction of wheat yield and quality characteristics, reveal wheat seeds forming ,nitrogen nutrition absorption, utilization, distribution patterns and explore in high-yield wheat and achieve the best possible quality of fertilization of an initial set of wheat nitrogen nutrition molecular genetic mechanism for wheat "three high" cultivation and provide a theoretical basis for breeding, we study wheat yield characteristics, quality characteristics,the law of absorption and utilization of nitrogen ,the development laws and accumulation dynamics of high molecular weight-glutin subunit(HMW-GS),the different expression of gladins and gene under the different nitrogen concentration by SDS-PAGE electrophoresis,A-PAGE electrophoresis and DDRT-PCR techniques in vitro cultivation.Research shows that :(1)Nitrogen concentrations in wheat yield index with a parabolic relationship. Nitrogen concentrations increased from 0% to begin, wheat yield has increased and their characteristics, but when nitrogen concentrations increased to 0.06%, wheat yield but a downward trend. The experiments found nitrogen concentrations affect yield mainly through affect the rate of grain,the weight of throusand seeds,the numberes of flowering and the lenghth of node.(2) The protein content of wheat seeds increase with the increasing concentration of nitrogen. nitrogen concentrations major impact globulin, Gladins in and glutenin, but the albumin almost no impact.(3) Combination of wheat seeds yield and quality characteristics, nitrogen absorption, the rate of use and transit patterns, and the principles of high-yield, high-quality and high-efficient ,the best nitrogen concentration is 0.06%.(4) Different nitrogen concentration does not affect the composition and the sequence of HMW-GS, but the time of HMW-GS formation and the volume of accumulation are related to nitrogen concentration, especially subunit 2. The study found that the interaction between subunit 2and subunit 12 by correlation analysis and multiple linear regression analysis.(5) Nitrogen concentration affects the volume and the time of formation of Gladins. Gladins content increase with nitrogen supply increased .When nitrogen concentrations is up to 0.12%, there are 3 unique belt ,respectively inωzone andλzone. The three unique belt content with the increased nitrogen concentrations increase.(6) We found 251 differences expressed belt, including 126 unique belts,68 stronger belts,57 lower belts. Two of which carried out cloning, sequencing and compared to the homologous sequence, and results showed that the identities of one is 86% with Triticum aestivum anther-specific gene promoter (raftin1b ),and the other Identities is 99% with Escherichia coli CFT073(Hypothetical protein yphG).
