Atropa belladonna L. is one of nightshade which is one of perennial herbage and belongs to herbol medicine. The frond of Atropa belladonna L. has some belladonna, hyoscyamine and scopolamine. In clinic it often be used for sedation, narcosis, acesodyne, antispastic, decrease glandulose secretion and enlarge pupil. 1990s, prof. Yang guodong applied scopolamine to be off the habit and get remarkable curative effect.Now, the research of tissue culture and breeding about Atropa belladonna L. is rare domestic. In the study, the systemic tissue culture of Atropa bennadonna L. is studied, which can be used for industrial production of Atropa belladonna L. and the further study; Simultaneously, based on tissue culture and used chromosome engineering, the technology and method on selectively breeding Atropa belladonna L. is studied in order to obtain the optimum polyploidy breeding method.The result in tissue culture of Atropa belladonna L. shows that the best sterile method is that 10min marinating by neutral scour, 30min overwashing by tap water, 6min sterilizing by 0.1% mercuric chloride, rinsing 3~5 times by sterile water. The bud induced rate is 100%, and the polluted rate is zero. And the result shows that the appropriate medium for inducing the differentiation on bud of the first culture about Atropa belladonna L. is MS+0.7mg/L 6-BA+0.2mg/L NAA+sucrose3.0%; The appropriate medium on step-generation and propagation of bud is MS+ 0.5mg/L 6-BA + 0.2mg/L NAA+sucrose3.0%, It is difficult to root for the first-generation bud of Atropa belladonna L., the appropriate medium is MS+0.2mg/L IBA+sucrose2.0%, the rate of rooting is 100%; The experiment of rooting about Atropa belladonna L. also shows that the times of step-generation and the days of step-generation affect the rooting of Atropa belladonna L. cultured sprout. As the times of step-generation exceeds 10 times, rooting is very easy on MS medium, the days of step-generation culture is 15~20d , rooting is easy comparatively; As it is 20~55d, rooting becomes difficult; But when the time is 90d, rooting becomes easily again on MS medium.On the base of Atropa belladonna L. tissue culture, the inducing polyploidy of Atropa belladonna L. is done by the solution of colchicines. Two methods are adapted: dipping and composite culture. The trial used for dipping has two kinds: stem-tip and axillary bud. The result shows that the dipping method is better than composite culture and the appropriate stuff is axillary buds. The highest inducing rate is 45% when the axillary buds are dipped in 0.10% colchicines...
