Soluble solid content is one of the important quality characters in tomato (Lycopeersicum esculentum.Mill) , which can evidently affect nutrition value, flavor and substance output of Tomato. It also has significant influence in cold and adversity resisting, fruit storing and frostbite preventing during transportation.Utilizing the genes increasing fruit soluble solid content from wild tomato is significant to improving fruit quality of cultivated tomatoes. Therefore, comprehensive researches have been done both home and abroad. Until now 38 QTLs related to fruit soluble solid content have been characterized and (QTL-NILs) or sub-nearly isogenic lines(sub-NILs) of this major QTLs have been developed. For instance, TA1218 is a introgression line of wild comate tomato (L.hirsutum ) LA1777, containing a 1.2cM fragment at the bottom of chromosome 1 of LA1777 in the background of E6203. QTLs analysis demonstrated that this fragment could significantly increasing fruit soluble solid content.Based on the previous study, TA1218 and its backcross offspring were employed to research into molecule assistant selection (MAS). Differential expression of the genes at different development stages of fruit was studied through cDNA-AFLP approach. The objectives were: to induct target gene into existing cultivated primary system and eliminate the influence of ill gene, then to create new breeding germplasm resource; Genes expression and function information at different development stages and the mechanism of accumulation of fruit soluble solid content were studied by identifying differential expression of the genes and separating the TDF related to fruit soluble solid content along with analyzing biologic information. The results were as follows:1. Four RELP marks were studied by CAPS translation analysis. TG161 and TG2372 were translated into CAPS marks, showing that RFLP marks can be RFLP marks can be converted into CAPS marks which can be easily manipulated with high efficiency and low cost.. The results possessed high value in quick genetic analysis, especially in genetic identification of mass sample in molecule breeding.2. 72 Ecor I/Mse I primers were used to select AFLP primers for TA1218 , E6203 and their F1 offspring but no primer is up to the standard of AFLP.3.
