Grain filling insufficiency is the major reason that cause low seed set percentage ,poor seed set and others traits closely related to bad grain quality in hybrid rice,especially in hybrids between indica and japonica rice or new plant type rice.Plant grain filling process is the coaction of plant source, sink and path. In this course plant source tissues(such as mature leaves) produce carbohydrates by the fixation of carbon dioxide during photosynthesis, then the surplus carbohydrates are exported to sink tisues(the roots, reproductive structures, developing organs and storage tissue)through the phloem in the form of sucrose. And sucrose transporter has been demonstrated to have the function of transferring sucrose through cytoplasm membrane.In this paper , two rice sucrose transporter(OsSUT2 and OsSUT5) gene had been introduced into rice restoring line Minhui86 ( Indica Saliva L . ) by agrobacterium-mediated transformation . Investigating on sucrose content,expression of this two genes in RNA level .activity of sucrose synthase and some indexes closely related to grain yields and quality , we finally attempted to discuss the potential function of this two genes in the rice. This results were summarized as following :1. Forward expression structure of OsSUT2 and OsSUT5 gene were introduced into rice restoring line Minhui86 ( Indica Sativa L . ) respectively by agrobacterium-mediated transformation, at last we obtained 106 and 68 independent clones of this two gene structure. Genetic analysis and x2 detecting showed that 27.8 % and 51.0% of T1 generation of this two gene structure accorded with Mendelian segregation law (3:1).2. PCR and sequencing showed that forward expression structure of OsSUT2 and OsSUT5 gene have been integrated into rice genome respectively.3.
