Papaya ringspot virus ( PRSV ) is an important seed-bome virus infecting cucurbitaceous crops but the standard detection of PRSV is absent. It's necessary to set up detection progresses of PRSV.In this study, DAS-ELISA (Double Antibody Sandwich ELISA) was used to detect PRSV in the leafages of the samples, which were collected in Haikou city, Fusui city, Zhengzhou city, Zhangzhou city. The gene was amplified by PCR with the cDNA reverse-transcript from the RNA of the leafages. The most suitable conditions with 56 °C and 2 U Tag DNA Polymerase were obtained by grad-PCR. Then DAS-ELISA and RT-PCR were employed to detect the PRSV in the seeds of pumpkin and the leafages collected from the plants.The results will provide rapid detection program of PVSV with RT-PCR.
