Alfalfa, which is a kind of perennial leguminous caudex, is the best and most important leguminous grass in the world . In 1972 ,Saunders, American scholar ,firstly gained the regenerative plants, which have accelerated the research in its field. It has been developed greatly for transgenic alfalfa , especially in setting receptor system, researching transform technologies, improving important agronomic characters.It is popular to develop tansgenic plants vaccines as soon as that new ideas is advanced, which make it possible for us to product cheap vaccines. Comparing with normal and other new technique vaccines, transgenic plants vaccines have much more indubitable superiority. It has been confirmed that it is possible for heterogenous antigen albumen to express in plants' cells. Also, the expressed antigen albumen in plants can cause the immunoreaction of animals and people. So , it is really feasible for producing vaccines with transgenic plants.The experiment is aiming at setting a high frequency regeneration system in virtue of alfalfa's tissue culture, then translating pig's RV'S VP7 gene to the high frequency regeneration alfalfas. Of course , it is basic of alfalfa's reactor that steady expression trans-gene alfalfa is obtained.GongNong No 1 , GongNong No 2 ,Alfalfa, The king of American alfalfa and Glassland Nol are used as the experimental materials .Cotyledons and hypocotyls of the mature seeds bourgeon are used as receptor materials. Then, induce cotyledons and hypocotyls making use of different type culture mediums, furthermore , investigate the important factors that influence explants to induce callus. As a result, callus can be induced from all kinds of the alfalfas, however , the rate of inducing is up to genotype, deference medium, concentration of hormone. For deferent types alfalfas' inducing, the inductivity of improved SH medium which has the highest rate of inducing embryogemic callus is better than MS medium, and the worst is B5 medium. But for 2,4-D,when its concentration is 2mg/l, all kinds of alfalfas have a high inductivity. At the same time , when cane sugar is used as the source of carbon, it is found that there is no diversity among different types ,and 30g/l is the appropriate concentration to induce and subculture as well as 10g/l is the appropriate concentration to take root. Add KT1. 0 mg/L and NAA1. 0 mg/L for the differentiate medium, the differentiate rate is 85.1%.
