Glucose Oxidase (GOD) oxidateβ-D glucose intoδ-lacttonic glucose with O2 , hydrolyze into acid radical glucose and H2O2 ulteriorly.H2O2 act as the secondary messenger in the action of plant disease resistance,it can sterilize and restrain pathogen straightway.The pMD18-T/GO was digested by HindⅢ,BamHⅠrestriction endonuclease, and cut down the target gene GO.The plant binary vector pROKII was digested by BamHI/SmaI.Then the target gene GO was ligated with the digested plant expression vector pROKII by T4 ligase.The recombinant vector pROKII/GO was confirmed by PCR amplification.The pROKII/GO was transferred into Agrobacterium LBA4404 competent cell by freeze and melt method and identified.The potato species Zihuabai,Tianjinkexin NO.1, Favorita were transformed with target GO gene by Agrobacterium mediated gene transfer,and through PCR amplification confirmed the GO gene was transformed of The potato species Zihuabai,Tianjinkexin NO.1.
