Peanut varieties Fenghua 2 and Luhua 11 were used for the study on improving genetic transformation efficiency mediated by Agrobacterium tumefaciens from three aspects. Firstly, we design the concentration and proportion of BA and NAA in the buds growing medium in the purpose of improving seedling percent, thereby pefectiong the accepted system of peanut genetic transformation mediated by Agrobacterium tumefaciens.Secondly, the screening systems of screening-medicament of marking gene Bar, Phosphinothricinacetyltransferase gene, and antibiotics of marking gene Npt II, Neomycin phosphotransferase gene, were clear. Thirdly, research on appropriate assemble of co-cultivation time and bacterial concentration in the infection process were done, for the purpose of increasing gene transformation efficiency.Technical procedure of peanut gene transformation mediated by Agrobacterium tumefaciens was optimized through research above. Fenghua 2 transgenic plants were obtained by the procedures. Polymerase chain rection of transgenic plant total genome results prove that gene Bar and geneγ– tmt,γ-Tocopherolmethyltransferase gene, were integrated into peanut genome.1.
