In the research the mechanism of botanic apoptosis is discussed by induced cells with extraneous source chemistry signals, the apoptosis related gene DAD1 in super-hybrid rice is cloned, and its prokaryotic expression vector which can express the DAD1 gene in E. coli. is constructed.1. Inducing the cell by extraneous source signals:a. different super-hybrid rice suspension cell systems are established by using callus tissue cells of different super-hybrid subculturing, the growth status of different suspension cells are compared, the different extracellular POD activity changes of different super-hybrid rice suspension cell when incubating are also discussed. The results show that super-hybrid rice suspension cell can be established easier when it is more dispersive or less brown. In the latter generations of rice suspension cell, the activity of extracellular POD and the cell suspensibility curve raises at first and then declines. The activities of extracellular POD induced by exogenous H2O2, CaCl2 and VK3 are different and the same concentration of VK3, CaCl2, H2O2 can all inhibit extracellular POD activities, especially to the callus tissue subculturing for six times. And the inhibitation of H2O2 is the most obvious. But the inhibitation reduces when H2O2 concentration and processing time adds. The extracellular POD activities enhance as the concentrations of VK3 and CaCl2 add when processing time between 6h and 12h. But the POD activities reduce when their concentration raises when processing for 48h. Then we conclude that the 3 chemicals all inhibit extracellular POD activities and induce cell apoptosis.
