The wool is the sheep production important economical character, the wool growth is controlled by many genetic factors, its quality and production is decided by sheep skin structure and hair follicle characters. Wool quantity and quality has obvious difference among different species, even different parts of a sheep, wool quantity and quality also has very significant difference, the major cause of the difference is gene differential expression.The study sampled Chinese merino sheep as experimental animal, applied mRNA differential display technique to separate differential expressed gene in sheep shoulder, abdomen or leg. The study employed 3 anchor primers and 8 random primers to proceed PCR amplification. PCR products were analyzed by 10% non denatured polyacrylamide electrophorisis, and the differential displayed genes were reclaimed as templates of second PCR amplification. Then the genes fragment were reclaimed again,The purified genes were cloned by pGEM-T vector and verified by Northern blot analysis, Finally ,the differential expressed genes were localized on sheep chromosome by In situ hybridization.Two gene fragments were obtained in the study, seqI was differential expressed gene fragment in sheep shoulder and abdomen and was localized on sheep chromosome 7; seqII was differential displayed gene in sheep leg and was localized on sheep chromosome 15.
