The Xinjiang Wapiti belongs to the second class protection wild animal in China, the function of antler in the medical, human healthy etc. is remarkable, the products are favored extremely, and is one of the resources worth developing and utilizition. The present study worked on the dissection jof antlers tissue structure and ultra-microstructure of antler periosteum, and have cultured and established antler stem cells in Wapiti. The results are as follows:1. The contents of collagen fibre is abundantly in antler, cell's quantity is low, maldistribution, not many kinds of cells, lie in periosteum mainly, cell take the form of long shuttleless loom, nuclear volume is high, kernel is obvious, some pair of kernels to be specifed, there are few cell devices, accord with the characteristic of the adult stem cell.2. The Wapiti 's ear skin fibroblasts cell and Wapiti antler stem cell culture methods and the culture system were developed.3. The result of antler stem cell culture the medium selection show that hDMEM, adopt FCS and NCS 1: 2 mixture is better than only one content. It should add 20% of the serum to primary cultrue, it is suitable for 10% serum to passage, it contributes to the growth of the antler cell to add the growth factor of FGF, androgen, insulin, adding 2.5ug/ml the androgen can promote cells to grow, the antler cell is right to inoculate the density and relatively sensitive, suitable for 106 / milliliter, too high or too low can cause cells apoptosis and feebly and split up.4. The antler stem cell has characteristic of sticking to the wall to grow, is taking the place of and culture early originally, in order to become the fibre cell kind spaciously and flatly mainly, taking the form of swirl while growing intensivly or bird nest form, reach the 3rd generation to present aging and differentiation, under the condition of culture in vitro, can become the bone cell and cartilage cell before dividing spontaneously.5. The antler cell adopt cell routine DMSO frozen methods is superior to program frozen methods and ethylene glycol cell routine freeze methods.6. It is positive that antler stem cell line established is dyed by AKP, the result from culture the shape, ultra-microstructure.
