As a member of Brassica, Brassica chinensis L. was a representative cross-pollinate vegetable crop, obvious heterosis was possessed. CMS was an important material used for heterosis.The studies carried on cytoplasm CMS researched to Brassica chinensis L, on the basis of establishing mtDNA and gDNA's isolation system , an optimized PCR reaction system for RAPD analysis was established: 25 primers selected from 200 primers amplified 144 RAPD fragments of mtDNA and 78 bands that occupied 54.2% of the whole amplified showed polymorphisms, each primer could amplify 4 to 9 polymorphic bands, with an average of 6.26 bands, amplified fragments ranged from 270 to 3000 bp; 20 primers selected from 200 primers amplified 107 RAPD fragments of gDNA and 49 bands that occupied 45.8% of the whole amplified showed polymorphisms, each primer could amplify 1 to 10 polymorphic bands, with an average of 5.35 bands, amplified fragments ranged from 270 to 10000 bp.The orf224 gene was obtained from cytoplasmic male sterility line of Brassica chinensis L. ,which was 691 bp. Sequences of comparison by Blast revealed that the gene homology between the DNA fragment from Brassica chinensis L. and orf224 from Brassica napus were 99%, proved that CMS of Brassica chinensis L..was caused by Pol CMS, rooted from mtDNA.
