Striped flea beetle, Phyllotreta striolata (Fab.), is a significant pest of cruciferous crops. It is spread widely across China and causes huge economic loss every year. The control and management of striped flea beetle has become the focus of various studies. In order to support the control measures with necessary theories, in this paper, the genetic diversity and differentiation of eight populations of striped flea beetle from different sites in Guangdong, Guangxi, Fujian, Jiangxi, Hunan, Guizhou, Yunnan and Sichuan provinces were analyzed by microsatellite analysis, for which five microsatellite markers were selected, and the results were as follows:1. The eight populations showed at their microsatellite loci highly polymorphism: the polymorphism information content (PIC) of 8 populations, Guangdong, Guangxi, Fujian, Jiangxi, Hunan, Guizhou, Yunnan, and Sichuan, were 0.8632, 0.8865, 0.8935, 0.9073, 0.8916, 0.9158, 0.9128 and 0.9074, respectively. Among the populations, that of Guizhou displayed the highest genetic diversity, with the value of its average heterozygosity (H) being 0.9213, while the population from Guangxi showed the lowest, with its value of H being 0.8768.2. Nei's genetic distances between the 8 populations were measured and the results showed that genetic distances between Fujian and Guangdong, Fujian and Jiangxi, and Sichuan and Guizhou populations were 0.1373, 0.1429 and 0.1429, respectively, implying no significant differentiation in their genetic structures; the values of genetic distance between Sichuan and other populations were relatively higher, which implies higher genetic differentiation between that population and the others. Differences in geographical distance, ecological environment and climatic condition may contribute to the result.3.
