It is hard to obatain regenerated plants from plant somatic hybridization. There are few reports about success of remote somatic hybrid plant before this research. It is only a start to study on how to increase the content of efficacious component in Chinese medicinal herbs by the method of somatic hybridisation. For example, it was only reported in the hybrids between P.quinquefolia L. and D.carota.In this work, somatic hybridization between Swertia tetrapteraand and its phylogenetically remote (interfamily) Bupleurum scorzonerifolium Willd was studied. The aim was: to establish an efficient system for hybrid plant regeneration; to investigate the effects of two parents relationship and UV dose on plant regeneration, phenotype and elimination and fragmentation of donor chromosomes; to analyze the differences of effective constituents and methanol-soluble components of regenerated plants between different phenotypes.The main process and results of this research were as follows: Protoplasts isolated from cultured cells of S. tetraptera as a donor were irradiated by UV at the intensity of 380uW/cm~2 for Os, 30s and 1min respectively and were fused with those of B. scorzonerifolium as a recipient by PEG method. Interfamily regenerated clones and plants were produced in different fusion combinations.1) In the symmetric and asymmetric hybridization between B. scorzonerifolium and S. tetraptera, 45 regenerated clones were produced in symmetric fusion combinations Ⅰ , 22 regenerated clones were obtained in asymmetric fusion combinations Ⅱ ,19 clones were in combinationsⅢ. Clone Ⅰ —3 and Ⅲ—4 differentiated into green plants. Regenerated plants from clone Ⅰ —3 clustered, which were phenotypically similar to S. tetraptera. The leaves of regenerated plants from clone Ⅲ—4 were narrow and long ,which were similar to B. scorzonerifolium.2) The clones were tested for hybridity by using morphological , cytological,...
