Objective:To study the inhibitive effect and mechanism of Matrine(Mat)on HeLa cell in vitro.Methods:Matrine at different concentration(0.5,1.0,1.5,2.0g/L)was used to cultivate HeLa cells in vitro.Mat’s inhibitive effect on HeLa cells was assayed by MTT after cells were cultivated for24h,48h and72h respectively;HeLa cells were treated by Matrine at different doses for48h,and the expression of eIF4E,4E-BP1,mTOR,ERK was detected by Western Blot analysis;1.0g/L Matrine were used to cultivate HeLa cells certain time,and the expression of p-eIF4E,p-4E-BP1,p-mTOR,p-ERK was detected by Western Blot analysis.Results:(1)The proliferation of HeLa cells was significantly inhibited by Matrine with dose and time related effect.Treated for certain time,the ratio of proliferative inhibition of Hela cells rised as the concentration of Matrine increased(F=235.04,P<0.01);At the same concentration,the ratio of proliferative inhibition of Hela cells rised as treating time prolonged(F=235.04,P<0.01).(2)The concentration of Matrine and its’treating time perform an mutual interaction on HeLa cells’proliferation inhibition rate(F=5.22,P<0.01).The ratio of proliferative inhibition of Hela cells maximized when trated by2.0g/L Matrine for72h(65.30±2.17,P<0.05).(3)In HeLa cells treated by Matrine at different concentration for48h,the expression of eIF4E,4E-BP1,mTOR,ERK have no difference controlling with the control group(F=0.17,0.93,0.11,0.20,P>0.05).(4)When treated by1.0g/L Matrine for different time,the expression of p-eIF4E,p-4E-BP1,p-ERK was significantly lower than that in the control group(F=43.48,107.23,143.27,P<0.05),while p-mTOR changed little(F=0.49,P>0.05).Conclusion:Matrine significantly restrained the proliferation of HeLa cells in vitro with time and dose dependent.It correlated with the down-regulating of p-eIF4E and p-4E-BP1,which were likely regulated by MAPK signaling pathway.
