Objective:According to the research of bone matrix’s structure and function. theresearch aims to synthesize polypeptide(RADA16-NBD) which has dual functions onosteoblasts, and to detect its effect on the migration、proliferation of inflammatory osteoblasst,thus lays foundation for the application of amino polypeptide in biology and clinical research.Polypeptide(RADA16-NBD)is synthesized By the method of changing the amino acidsequence and on the foundation of RADA16.Methods:Resuscitate osteoblast cell lines and subculture; Prepare hydrogel RADA16-NBD by mixing method and observe its structure by scanning electron microscopy. At the sametime inoculate inflammatory osteoblasts on peptide hydrogel bracket RADA16-NBD; Observethe migration ability of osteoblast by the method of scarification; Detect the effect of peptidebracket on the adhesion and proliferation ability of inflammatory osteoblast cells by usingCCK-8.Result:Through scanning electron microscopy, it is seen that cells get good extension inhydrogel. CCK-8shows that inflammatory osteoblasts have good adhesion and proliferationability in hydrogel RADA16-NBD. The scarification experiment shows that hydrogelRADA16-NBD have a strong effect ion on the migration ability of inflammatory osteoblasts.Conclusion:Self-assembling peptide hydrogel can support the adhesion、spreading、migration and proliferation of inflammatory osteoblasts, and shows its strong effection. Its dualfunction in osteoblasts and inhibiting the activation of the ofinflammation is indicated, thus pavea new way of its application in the treatment of inflammatory bone resorption, especially inbone defects caused by periodontitis. This experiment lays foundation for the the followinganimal experiment.
