objective To investigate the relationship between TLR4gene polymorphisms and the etiological subtypes of ischemic stroke and liability of atherosclerosis(AS) in different vascular bed in the Han population in north of china.Methods Our study included389ischemic stroke patients (286for LAA and103for SAO) as well as300controls. LAA group was divided4subsets according to angiostegnosis condition (including the number of vascular stenosis; intracranial/extracranial aterial stenosis; combined peripheral artery stenosis like renal artery, coronary artery and artery of lower extremity; anterior/posterior cerebral circulation stenosis). Polymerase chain reaction-restriction fragment length polymorphism or pyrosequencing was performed to analysis3SNPs of TLR4. The software package Haploview was used to analysis haplotype.Results Genotypic effects were estimated using Logistic regression analysis adjusted for age, gender, Hypertension, Diabetes, Coronary Disease, smoking status, alcohol consumption and blood fat level. The TLR43SNPs (rsl927914, rs1927911,rs2149356) had no association with SAO group (P>0.05). The gene polymorphisms of rs1927911and rs2149356had association with LAA stroke. Haplotype analysis was revealed a significant different expression between LAA group and controls. Compared with controls, the frequency of haplotypes H2and H8was lower (OR=0.439,95%CI=0.306-0.631, P=5.92e-006;OR=0.332,95%CI=0.181-0.608, P=0.000) and haplotype H3was significant higher in LAA group (OR=2.301,95%CI=1.705-3.106, P=3.40e-008).Conclusion Our findings show TLR4SNPs polymorphisms had association with LAA stroke subtype in the Han population in north of china, However no association with Atherosclerotic Lesion in Different Vascular bed.
